چکیده

MSc Thesis Abstract Background: The use of vaccination in the control schistosomiasis at a population level induced low cure rates and also resistance to drugs has been recently recorded Egyptian patients. For these reasons, search for new with adjuvant anti-schistosomal activity is urgently needed. Aim study: study focused on newly synthesized ligands that play central role as inducers innate adaptive immune responses are therefore considered potential targets vaccine adjuvants. These include Polyinosinic: polycytidylic acid (Poly (I:C) (TLR3L) and  Zymosan (TLR2L) which have displayed an impressive array biological activities including antitumor antiviral effects. present investigated their effects S. mansoni-infected mice. Materials Methods: was divided into two experimental settings treatment settings. In settings, mice were assigned 8 groups. Mice vaccinated 2 times weeks intervals, then infected mansoni. Five post-infection, euthanized analysis. non-vaccination settings,  determination worm burden, tissue eggs count, assessment liver function tests, parameters oxidative stress done. Results: Vaccination by Poly increased counts platelets, segmented neutrophils, eosinophils, monocytes while decreases lymphocytes indicating immunological effect ligands. current showed soluble antigen protein (SWAP) associated improvement enzymes compared SWAP alone. apparent granuloma volume. TLR had no side normal healthy reduced adverse caused mansoni through enzymes, stress, reduction ova count.  Conclusion: This first assesses adjuvants infection. Our recommendation increase time both significant preserve parameters. (This thesis approved Faculty Science, Tanta University, Egypt November 26, 2012).

Immunopathology of Schistosoma mansoni infection.

Schistosomiasis mansoni is a chronic helminthic disease that affects about 100 million people in the tropics. The worms have a life span of 5 to 10 years, and they live in the mesenteric veins of the host. Lightly infected individuals are asymptomatic or manifest mild intestinal symptoms. Heavily infected individuals often develop severe morbidity with hepatosplenomegaly, sometimes with a fatal...

Immunization against Egyptian Schistosoma mansoni infection by multivalent DNA vaccine.

The development of multivalent vaccines consisting of several antigens is a novel approach to creating broad-range protection against different parasite strains and parasite life cycle stages. We have previously confirmed that the schistosome Sm21.7 and SmFimbrin (SmFim) proteins could induce protection in mice. Therefore, this study aimed to construct the multivalent DNA vaccine Sm21.7-SmFim/p...

Schistosome Syntenin Partially Protects Vaccinated Mice against Schistosoma mansoni Infection

BACKGROUND Schistosomiasis is a neglected tropical disease caused by several species of trematode of the genus Schistosoma. The disease affects more than 200 million people in the world and causes up to 280,000 deaths per year, besides having high morbidity due to chronic illness that damages internal organs. Current schistosomiasis control strategies are mainly based on chemotherapy, but many ...

Potential immunomodulatory effects of plant lectins in Schistosoma mansoni infection.

Lectins are sugar-binding glycoproteins that can stimulate, in a non-antigen-specific fashion, lymphocytes, leading to proliferation and cytokine production. Some lectins are utilized as in vitro mitogenic lymphocyte stimulators and their use as immunomodulators against infectious diseases has been evaluated experimentally. In the experimental murine model, the immune response to schistosomiasi...

Fasciola gigantica Fatty Acid Binding Protein (FABP) as a Prophylactic Agent against Schistosoma mansoni Infection in CD1 Mice

Although schistosomicidal drugs and other control measures exist, the advent of an efficacious vaccine remains the most potentially powerful means for controlling this disease. In this study, native fatty acid binding protein (FABP) from Fasciola gigantica was purified from the adult worm's crude extract by saturation with ammonium sulphate followed by separation on DEAE-Sephadex A-50 anion exc...
